If you’ve ever been told you can only absorb 30 grams of protein at a time, or that skipping past 24 hours of fasting requires nothing but water, this episode challenges both ideas with real research.

I am joined by Vanessa Spina of Ketogenic Girl and the Optimal Protein Podcast to unpack a landmark human study on protein, fasting-mimicking diets, and autophagy, and to walk through exactly how a protein-sparing modified fast works in practice.

The Truth About Protein Absorption and Timing

Vanessa and I start with the isotopically labeled protein study out of the group led by Dr. Luc van Loon, which tracked amino acids from a 100-gram protein meal in trained males for 12 hours (well beyond the four hours prior studies had measured) and found the amino acids were still being taken up into muscle tissue well past the old “30-gram limit.”

The takeaway: total daily protein intake matters far more than perfect timing or an “anabolic window” right after a workout.

Why High Protein Doesn’t Block Autophagy

The core of the episode is a 2025 University of Nottingham study that put 46 healthy adults on a seven-day, 850-calorie fasting-mimicking diet, either low-protein (10% of calories) or high-protein (30% of calories). Both groups lost weight and fat, dropped fasting glucose and IGF-1, and showed upregulated autophagy gene expression.

But the high-protein group came out ahead: it saw a significant drop in visceral fat, lower triglycerides, better heart rate variability, better gut microbiome diversity, and activation of the autophagy-initiation gene ULK1 — none of which showed up to the same degree in the low-protein group.

Putting It Into Practice: The Protein-Sparing Modified Fast

Vanessa shares how she used a protein-sparing modified fast (PSMF) — roughly 800 to 1,000 calories a day, high protein (around 1 gram per pound of body weight), very low fat — to lose 14 pounds of fat while preserving muscle, then shifted to one PSMF day per week for maintenance.
